I'm running it on 12 volts. Model trains and track switches are controlled in n scale.

Nominal three. 5 amps? No. Although it could withstand such a high spike, it is not rated for such a high draw.

Is it possible to power four 12 volt dc motors with a current of one amp each? 5a each, powered by a 12-volt battery capable of providing 2-hours of power ?

That does not appear to be the case. You have only 3a to work with, but you require 6a. I'm not an expert in electronics, so all I can do is guess.

Is it possible to stack this board on top of a motor shield made by Adafruit?

I've used these in place of the Adafruit motor control shield (the v2 version, which this is essentially a knockoff of). The address jumper will need to be soldered to give the second board a unique address, but that should be it.
